Toxicity assessment of PM2.5 brake wear emissions using aerosol-based exposure models with A549-HMEC cell co-culture

Preliminary test of toxicological responce of brake wear particles using Vitrocell Cloud Alpha 12 system under airliquid interface (ALI) conditions, mimicking more realistic exposure conditions to airborne PM2.5.

  • In the co-culture the formation of a functional alveolar-capillary barrier is assessed using IF staining of epithelial cells of Type II (Hopx) and ciliated cells (acetylated tubulin).
  • A homogeneous, tight cell layer is observed, with a thickness ranging from 10 to 15 µm for A549 cells.
  • Nebulization of brake wear particles is performed using the Vitrocell Cloud Alpha 12 system, resulting in a uniform distribution of particles across test inserts.
