Testing of conventional and e-cigarettes: using EpiAirway and EpiAlveolar with the VITROCELL® in vitro Exposure System

New MatTek application note shows a state-of-the-art in vitro set up by VITROCELL® Systems for testing combustion cigarettes as well as electronic cigarettes with the endpoints toxicity, oxidative stress, TEER, beating cilia, gene expression and mucus secretion.

Abstract
The installation consists of the VITROCELL® VC1 Smoking Machine, a dynamic VITROCELL® Dilution System and the VITROCELL® 12/6 CF Exposure Module. It is well suited for testing whole smoke from conventional cigarettes as well as vapours from electronic cigarettes.

Quantitative PCR of CYP1A1 shows significant increases in expression after exposure of EpiAirway to TCIG smoke (48 puffs) and ECIG vapor (400 puffs).

8-Isoprostane in EpiAirway culture medium is significantly increased after both TCIG and ECIG exposure. 8-Isoprostane significantly increased after TCIG smoke exposure (48 puffs), or exposure to ECIG vapor (400, 200, 100 and 50 puffs).
