Big tobacco embraces in vitro toxicology

Manufacturers invest in nonanimal tests to evaluate health risks of e-cigarettes and other next-generation tobacco products

The complexity of next-generation tobacco products has increased by a number of factors since FDA was given responsibility to regulate tobacco. Testing e-liquids with animals is not practical, economically and not that accurate. The Vitrocell system generates airborne materials from test products, such as smoke from reference cigarettes and vapor from e-cigarettes, in a manner that simulates how a person would puff. This artcile shows, how IIVS scientists use human donor tissues, obtained from organs, to create cell cultures and in vitro test systems.

The experimental setup for the aerosol exposure consisted of a VC 10 smoking robot, a dilution system, and a Vitrocell 12/3 glass module the exposure of nicotine and TPM in the mainstream aerosols from 3R4F and novel tobacco vapor on MucilAir™ cells. The results are shown in cytotoxicity, IL-8, IL-1ß and gene expression.

The article shows the successfull qualification of the Vitrocell VC10 smoking robot. The robot was used to  expose bacterial cells to cigarette whole smoke generated from 3R4F reference cigarettes and Eclipse cigarettes. A comparison was made between whole smoke from a tobacco heating and a combustible cigarette.

The study shows an assessment of VC1 aerosol generation that was conducted by measuring puff-by-puff in vitro nicotine concentration of 3R4F cigarettes (ISO and HCI regimes), e-cigarette (CRM81 puffing profile), and THP (HCIm regime). The aerosols were applied to in vitro or ex vivo cellular and tissue systems that model the respiratory tract, such as reconstructed human airways (RHuA), which are grown at the air–liquid interface (ALI). Additionally a comparison between the Vitrocell VC1 (U.S. Laboratory) and the VC10 (UK Laboratory) Smoking Robot was done.

New Products at VITROCELL® User Group Meeting 2018

Following the VITROCELL® User Group Meeting 2017 in Baltimore, international scientists from Canada, Europe, Japan and USA reconvened in March 2018.

The informal event organized into short presentations and posters was followed by open discussions. It was an excellent opportunity to discuss the latest developments of VITROCELL®, to exchange your experience in working with the equipment and to meet other fellow researchers. The meeting took place prior to the international Society of Toxicology 57th Annual Meeting and ToxExpo, March 11 – 15, 2018, in San Antonio, USA - one of the largest international conferences related to toxicology.

Focus of the event was to share VITROCELL® activities since the last meeting, to exchange latest research results, to give an update on VITROCELL’s participation in major research programs as well as the presentation of new products for 2018.

This study investigated the in vitro mutagenicity of e-cigarettes and designed a study to deliver e-cigarette aerosol on an undiluted per puff basis under extreme testing conditions, using the Ames assay across a broader range of bacterial tester strains. The Vitrocell VC 10 smoking robot, previously used to generate and dilute cigarette smoke aerosols was modified to enable the testing of an undiluted e-cigarette aerosol.

A reconstituted 3D human airway epithelium was exposed to undiluted aerosols at the air-liquid interface, using a Vitrocell VC 10 under ISO and HCI conditions. A 3R4F reference cigarette and a commercially available e-cigarette product were tested on their effect on CBF and TEER.
