The purpose of this study is to evaluate oxidative stress induced by whole cigarette smoke based on the air-liquid interface exposure system. The cytotoxicity of whole smoke were measured by MTT and NRU assays, and several intracellular and extracellular oxidative stress biomarkers such as GSH and GSSG, MDA, HNE, EC-SOD and 8-OHdG were investigated in A549 and BEAS–2B cells.

PETA science consortium donates $400K in VITROCELL in vitro test equipment

London -- Today, the PETA International Science Consortium Ltd. announced that it is providing equipment to laboratories working on human-relevant methods to replace the use of animals in inhalation testing. Valued at more than $400,000, VITROCELL Systems inhalation exposure machines were awarded to four institutions in the U.S., U.K., and Belgium that are leading the field of in vitro (non-animal) inhalation toxicology. These donations complement the Consortium's recent workshops and webinars focused on replacing inhalation testing in animals.

The aim of this study was to test whether temperature and relative humidity has an influence on the cytotoxicity to BEAS-2B cells on a VITROCELL® 6 air-liquid interface system by measuring the viability (WST-1 cell proliferation assay) and lactate dehydrogenase (LDH) release.

Cytotoxicity, cell morphology, cilia beating frequency, mucociliary clearance, gene expression analysis of oxidative stress and (pro‑)inflammatory markers as well as (pro‑)inflammatory cytokine secretion were successfully observed.
